چکیده

The purpose of this study was to examine the ul trastructural relationships of cholesterol crystals and apatite deposits in human atherosclerotic lesions. Segments of human aortic atherosclerotic lesions were obtained at autopsy , fixed in gl utaraldehyde and dehydrated without using any organic solvents. The aortic segments were coated with carbon and subjected to various scan ning electron microscope analyses. These included secondary electron imaging , back scattering of primary electrons, energy dispersive X-ray analysis of selected spots followed by area mapping of calcium and phosphorus , and cathodoluminescence. The information gathered from scanning of selected areas in the lesions by all the techniques showed that cholesterol crystals and apatite deposits are close to each other, within 10 μm distance or less. Cholesterol crystals are often surrounded by or adjacent to apatite. The results indicate that cholesterol and apatite crystals form closely linked agglomerates in human ath erosclerotic lesions. Further studies are needed to determine whether precipitation of calcium and cholesterol are somehow linked during atherosclerotic lesion development.

Raman spectroscopy imaging reveals interplay between atherosclerosis and medial calcification in the human aorta

Medial calcification in the human aorta accumulates during aging and is known to be aggravated in several diseases. Atherosclerosis, another major cause of cardiovascular calcification, shares some common aggravators. However, the mechanisms of cardiovascular calcification remain poorly understood. To elucidate the relationship between medial aortic calcification and atherosclerosis, we charact...

Isolation and characterization of a complement-activating lipid extracted from human atherosclerotic lesions

The major characteristics of human atherosclerotic lesions are similar to those of a chronic inflammatory reaction, namely fibrosis, mesenchymal cell proliferation, the presence of resident macrophages, and cell necrosis. Atherosclerosis exhibits in addition the feature of lipid (mainly cholesterol) accumulation. The results of the present report demonstrate that a specific cholesterol-containi...

Resveratrol protects against diet-induced atherosclerosis by reducing low-density lipoprotein cholesterol and inhibiting inflammation in apolipoprotein E-deficient mice

Objective(s):Resveratrol (RES) is a polyphenol compound that has been shown a promising cardioprotective effect. However, some reports have yielded conflicting findings. Herein, we investigated the anti-atherosclerotic effects of RES in apolipoprotein E (apo E)-deficient mice on a high cholesterol diet. Materials and Methods: Firstly, atherosclerosis was induced by feeding a high cholesterol di...

Development of the atherosclerotic core region. Chemical and ultrastructural analysis of microdissected atherosclerotic lesions from human aorta.

Lipid deposits in human atherosclerotic fibrous plaques exhibit marked differences in chemistry and ultrastructure from lipid deposits in fatty streaks, leading some investigators to question whether fibrous plaques originate from fatty streaks. To examine lesion transition, we employed lipid microanalysis, electron microscopy, and immunohistochemistry on fatty streaks, fibrolipid lesions (smal...

Rabbit Atherosclerotic Lesions

Previous studies have demonstrated the presence of apoprotein (apo) E protein and message in arterial lesions. To determine the source of the synthesized apoE, we performed simultaneous in situ hybridization and immunocytochemistry on human and rabbit atherosclerotic tissue. Studies of serial sections of aortic atherosclerotic lesions from humans and hypercholesterolemic New Zealand White rabbi...
